Structural variant evolution after telomere crisis

چکیده

Abstract Telomere crisis contributes to cancer genome evolution, yet only a subset of cancers display breakage-fusion-bridge (BFB) cycles and chromothripsis, hallmarks experimental telomere identified in previous studies. We examine the spectrum structural variants (SVs) instigated by natural crisis. Eight spontaneous post-crisis clones did not show prominent patterns BFB or chromothripsis. Their crisis-induced rearrangements varied from infrequent simple SVs more frequent complex SVs. In contrast, chromothripsis occurred MRC5 fibroblast that escaped after CRISPR-controlled telomerase activation. This system revealed convergent evolutionary lineages altering one allele chromosome 12p, where short likely predisposed fusion. Remarkably, 12p events were stabilized independent fusions 21. The data establish can generate wide implying lack genomes does indicate absence past

Telomere identity crisis.

Cells are designed to be intolerant of breaks in DNA, yet it is critical that cells do not identify the ends of linear chromosomes, called telomeres, as damaged DNA ends. Telomeres therefore must somehow prevent the recognition and subsequent repair of chromosome ends as double-strand breaks (DSBs), although how this is achieved is poorly understood. Chromosomal breaks can occur as a result of ...

Chromothripsis and Kataegis Induced by Telomere Crisis

Telomere crisis occurs during tumorigenesis when depletion of the telomere reserve leads to frequent telomere fusions. The resulting dicentric chromosomes have been proposed to drive genome instability. Here, we examine the fate of dicentric human chromosomes in telomere crisis. We observed that dicentric chromosomes invariably persisted through mitosis and developed into 50-200 μm chromatin br...

Telomere Crisis and Chromosome Shattering in Cancer

Although cancer development involves gradual step-wise mutational alterations, recent work has pointed to an additional contribution of episodic massive chromosome shattering phenomena, including chromoplexy and chromothripsis. How chromoplexy and chromothripsis are initiated is unknown. We hypothesize that chromosome shattering originates from end-to-end fused dicentric chromosomes, which occu...

Alternative end joining, clonal evolution, and escape from a telomere-driven crisis

Telomere dysfunction and fusion play key roles in driving genomic instability and clonal evolution in many tumor types. We have recently described a role for DNA ligase III (LIG3) in facilitating the escape of cells from crisis induced by telomere dysfunction. Our data indicate that LIG3-mediated telomere fusion is important in facilitating clonal evolution.
